Effect of waste mango silage on the in vitro gas production, in situ digestibility, intake, apparent digestibility, and ruminal characteristics in calf diets

Preview Abstract
Mexico is the fifth largest producer of mangoes in the world. For the conservation of agro-industrial waste and crop residues, the ensiling technique has shown good results. This study aimed to evaluate the effect of increasing the level of mango silage (86% waste mango and 14% pangola grass hay) in calf diets on in vitro gas production, in situ digestibility, intake, apparent digestibility, and ruminal characteristics. The diets contained 0 (T0), 30 (T1), 45 (T2), and 60% (T3) mango silage. The partial (24, 48, and 72 h) and cumulative (72 h) biogas, CH4 production, and degradation were determined in the in vitro evaluation. In situ digestibility and estimators of fermentation kinetics of dry matter (DM) and organic matter (OM) were determined. Intake, apparent nutrient digestibility, and rumen parameters of calves (200 kg) were evaluated in a 4 × 4 Latin square design. Response to increased mango silage was calculated by linear and quadratic orthogonal contrasts. In vitro partial and cumulative biogas production decreased linearly (p < 0.05), and the partial and cumulative CH4 production did not show linear or quadratic contrast (p > 0.05); in vitro DM degradation, in vitro neutral detergent fiber degradation, and in vitro acid detergent fiber degradation showed a linear increase (p < 0.05). In situ dry matter digestibility (DMDis), in situ organic matter digestibility (OMDis), b, a + b, c, and effective digestibility (ED) of DMDis, a, a + b, c, and ED of OMDis increased linearly (p < 0.05). Dry matter intake, OM intake, and crude protein intake showed a linear increase (p < 0.05); NDF intake and ADF intake presented a quadratic behavior (p < 0.05). Apparent digestibility of DM, OM, CP, and hemicellulose, pH, N-NH3, total bacterial count, acetate, propionate, butyrate, volatile fatty acids, acetate: propionate ratio, cellulolytic bacteria, and protozoa did not present a linear or quadratic orthogonal effect (p > 0.05). The in vitro, in situ, and in vivo variables demonstrated that up to 60% mango silage can be used for the intensive fattening of calves in confinement. Preview Abstract
All serotypes of Salmonella enterica are considered potentially pathogenic. However, the non-typhoidal Salmonella (NTS) serotypes vary considerably in terms of pathogenicity and the severity of infections. Although diverse serotypes of NTS have been reported from tropical seafood, their sources, physiological characteristics, and virulence potentials are not well understood. This study aimed to compare the physiological characteristics of selected serovars of Salmonella from seafood and investigate possible variations in the distribution of known genes within the pathogenicity islands. A series of biochemical tests, including carbohydrate fermentation and amino acid decarboxylation tests were performed to physiologically compare the isolates. The genetic characterization with respect to putative virulence genes was done by screening for genes associated with Salmonella pathogenicity island (SPI) I– V, as well as the toxin- and prophage-associated genes by polymerase chain reaction. Irrespective of serotypes, all the isolates uniformly harbored the five SPIs screened in this study. However, some virulence genes, such as the avrA, sodC, and gogB were not detected in all Salmonella isolates. The biochemical profiles of Salmonella serotypes were highly conserved except for variations in inositol fermentation and citrate utilization. All the isolates of this study were weak biofilm formers on polystyrene surfaces. The pathogenicity profiles of environmental NTS isolates observed in this study suggest that they possess the virulence machinery necessary to cause human infections and therefore, urgent measures to contain Salmonella contamination of seafood are required to ensure the safety of consumers. Virome diversity of Hyalomma dromedarii ticks collected from camels in the United Arab Emirates

Preview Abstract
Viruses are important components of the microbiome of ticks. Ticks are capable of transmitting several serious viral diseases to humans and animals. Hitherto, the composition of viral communities in Hyalomma dromedarii ticks associated with camels in the United Arab Emirates (UAE) remains unexplored. This study aimed to characterize the RNA virome diversity in male and female H. dromedarii ticks collected from camels in Al Ain, UAE. We collected ticks, extracted, and sequenced RNA, using Illumina (NovaSeq 6000) and Oxford Nanopore (MinION). From the total generated sequencing reads, 180,559 (~0.35%) and 197,801 (~0.34%) reads were identified as virus-related reads in male and female tick samples, respectively. Taxonomic assignment of the viral sequencing reads was accomplished based on bioinformatic analyses. Further, viral reads were classified into 39 viral families. Poxiviridae, Phycodnaviridae, Phenuiviridae, Mimiviridae, and Polydnaviridae were the most abundant families in the tick viromes. Notably, we assembled the genomes of three RNA viruses, which were placed by phylogenetic analyses in clades that included the Bole tick virus. Overall, this study attempts to elucidate the RNA virome of ticks associated with camels in the UAE and the results obtained from this study improve the knowledge of the diversity of viruses in H. dromedarii ticks. Preview Abstract
Avian pathogenic Escherichia coli (APEC) is the causative agent of colibacillosis, one of the most prevalent bacterial diseases responsible for significant economic losses in the poultry industry worldwide. This study aimed to assess the antimicrobial resistance (AMR) patterns of APEC isolates recovered from poultry in east Algeria and estimate the prevalence of extended-spectrum β-lactamase (ESBL)-producing isolates. In the slaughterhouse of Batna City (Algeria), livers indicating colibacillosis were sampled from 204 suspected carcasses with growth retardation and generalized congestion. Escherichia coli isolation and identification were performed on MacConkey agar using conventional methods and the API 20E system. Antimicrobial resistance susceptibility was tested by the disk diffusion method according to the Clinical Laboratory Standards Institute Guidelines. Extended-spectrum β-lactamase detection was carried out using the double-disk confirmation test. One hundred sixty E. coli isolates were recovered (one isolate per sample). Avian pathogenic Escherichia coli isolates showed high levels of resistance to ampicillin and tetracycline (100%), nalidixic acid (95%), ofloxacin (93.75%), doxycycline (91.87), ciprofloxacin (87.50%), trimethoprim/sulfamethoxazole (62.50%), gentamycin (32.50%), chloramphenicol (27.50%), amoxicillin/clavulanic acid (16.25%), colistin (14.37%), and nitrofurantoin (10.62%). All strains were multidrug-resistant to at least three antibiotics, and more than half (52.52%) of the isolates were resistant to at least seven antibiotics. All isolates were susceptible to ceftriaxone, ceftazidime, and aztreonam. Two E. coli isolates were ESBL producers (1.25%). Avian pathogenic Escherichia coli resistance to most antimicrobial agents used in poultry may lead to antimicrobial therapy failure. Preview Abstract
Food poisoning caused by Salmonella enterica serovars is the most common type of foodborne illness. Tainted chicken meat is a major vector for spreading these serovars throughout the food supply chain. Salmonella isolates that developed resistance to commonly used antimicrobials pose a noteworthy risk to public health, yet there has been a lack of data on this issue in Iraq. Therefore, it is crucial to address these serious public health challenges with an adequate database on the occurrence and antibiotic resistance of these serovars. This study aimed to determine the frequency of occurrence of Salmonella enterica serovars Enteritidis and Typhimurium (S. Enteritidis and S. Typhimurium), antimicrobial resistance (AMR), and prevalence of multidrug resistance among S. Enteritidis and S. Typhimurium isolated from poultry meat collected in Wasit Province in Iraq. A total of 150 raw and frozen poultry meat samples were gathered from retail markets in various locales across the Wasit Governorate in Iraq. Salmonella spp. were successfully cultured and identified using the technique recommended by ISO 6579:2002, with minor modifications. The multiplex polymerase chain reaction approach was used to confirm Salmonella spp. (S. Enteritidis and S. Typhimurium). A disk diffusion test was performed to determine the susceptibility to particular antimicrobial agents, and 12 different antimicrobial agents were evaluated. Only 19 of the 150 (12.7%) samples tested positive for Salmonella (16% and 11% were isolated from raw and frozen chicken meat, respectively). S. Enteritidis accounted for 63.2%, whereas S. Typhimurium accounted for 36.8%. Nalidixic acid resistance was the most common (73.7%), followed by sulfamethoxazole-trimethoprim (63.2%) and tetracycline (63.2%), but gentamicin and ciprofloxacin (up to 15.8%) only had modest resistance. Antibiogram of S. Enteritidis and S. Typhimurium yield 13 antibiotypes. Among the 19 Salmonella isolates, 12 of 19 (63.2%) established resistance to no less than three categories of antimicrobials. This study highlighted the necessity of limiting the utilization of antibiotics in animal production by providing vital information regarding the frequency and AMR of Salmonella at markets in Wasit Province. Therefore, risk assessment models could use these data to lessen the amount of Salmonella passed on to humans in Iraq from chicken meat. Effect of short-term nutritional supplementation of green microalgae on some reproductive indicators of Anglo-Nubian crossbred goats

Preview Abstract
Despite the wide spectrum of uses, one of the chief drawbacks to expanding microalgae as a food supplement in livestock is the lack of a regimen protocol with established dosage and time length of supplementation. Therefore, this study aimed to investigate the effect of short-term supplementation with increasing doses of microalgae on ovarian response in goats reared in northeast Brazil. Twenty-eight goats had their follicular waves synchronized using three injections of a prostaglandin analog at 7-day intervals. Goats were allocated to groups that received daily oral Chlorella supplementation for 7 days, respectively: 5 g, GMA5 group (n = 7), 10 g (GMA10; n = 7), and 20 g (GMA20; n = 7). The control group (GMA 0; n = 7) received a drench of water. The groups showed a quadratic increase (p = 0.0156) in kidney fat thickness but there was a significant reduction in dry matter intake in the GMA20 group. The GMA20 group showed higher glucose levels and glutathione peroxidase (p < 0.05). There was a decrease in plasma cholesterol (p < 0.05) in the 10 and 20 g treatments. The number of total follicles increased quadratically. Follicles <3 mm increased linearly (p = 0.0113) for microalgal supply. The GMA10 and GMA20 groups had the highest values (p < 0.05) among the treatments. After inducing ovulation, there was a significant increase in follicles >3 mm in the GMA10 group, which also showed a greater (p < 0.05) area of intraovarian blood perfusion and pulsatility index of the ovarian artery. We conclude that for 7 days of supplementation, the administration of 10 g of microalgae appears to be the most efficient dosage for stimulating the ovarian response in tropical goats. Preview Abstract
Pelung cockerels (Gallus gallus gallusGallus gallus gallus [Linnaeus, 1758]) are different from other native cockerels in that they have a long and unique voice, in addition to their tall, large, and sturdy body with a relatively heavy body weight (BW). The sound quality of pelung cockerels is affected by the structure of the syrinx and their large and strong chest muscles. The performance of the chest muscles, and subsequently its voice, is influenced by the hormone testosterone. The shell of blood clams (Anadara granosa Linnaeus, 1758), a saltwater bivalve is known to contain a natural aromatase blocker (NAB) capable of blocking the aromatase enzyme from converting testosterone to estradiol. This generates consistently high levels of testosterone. This study aimed to determine the effect of blood clam shell powder (BCSP) as an NAB on the growth, pectoralis muscle performance, and testes of pelung cockerels. The study design was a completely randomized design, with 16 pelung cockerels aged 40–56 weeks divided into four treatment groups: T0 (control); T1 (BCSP [A. granosa] 0.9 mg/kg BW); T2 (zinc sulfate [ZnSO4] 0.9 mg/kg BW); and T3 (testosterone 3 mg/day). The animals were acclimatized for 7 days and then given dietary treatments for 56 days. The measurement of the comb, wattle, and chest circumference (CC) of pelung cockerels was performed on days 0, 14, 28, 42, and 56. At the end of the treatment, the pelung cockerels were sacrificed and the data of the pectoralis muscle weight (PMW), testis weight (TW), and area of the pectoralis muscle (APM) were measured. Samples of pectoralis muscle and testes were taken and fixed in 10% neutral buffer formalin for histology. The proliferating cell nuclear antigen (PCNA) was identified by immunohistochemical staining. To measure fascicle area (FA), myofiber area (MA), and enumerate, the fascicle myofibers (NM) histology preparations were stained with hematoxylin and eosin (H and E). Testicular preparations were stained with H and E to measure the diameter of the seminiferous tubules (DST) using ImageJ software. The growth performance on day 56 showed significantly (p < 0.05) higher differences of CC in T1 compared to T2 and T0, in T1 and T3 compared to T0, and in T3 and T2 compared to T0. Pectoralis muscle results, that is, FA, NM, MA, and PCNA-positive cells, showed that cockerels on treatment T3 had significantly higher results than other treatments, T1 was significantly different from T2 and T0, and T2 was significantly different from T0. In addition, the TW and DST measurement of cockerels on treatment T3 were significantly reduced (p < 0.05) than the other treatment groups. The oral administration of BCSP in the role of a NAB at a dose of 0.9 mg/kg BW for 56 days improved the growth performance and pectoralis muscle, especially the CC, FA, NM, MA, and PCNA-positive cells parameters, but did not affect the PMW, APM, and testis of pelung cockerels. The administration of testosterone at 3 mg/day for 56 days contributed to the decrease in TW and DST, as well as atrophy of the seminiferous tubules of pelung cockerels. Preview Abstract
Good management in goats is known for good quality health and increasing productivity. The physiological change studies in goats are limited despite some existing studies on the relationship of various patterns to growth rates. This study aimed to determine the hematological parameters, oxidative stress, and parasitic infection in three management systems in Thai native goats. A total of 18 male goats were randomly assigned to the three systems: The free-range model (FREE), the semi-intensive model (SEMI), and the kept-in-a-cage model (BARN) for 35 days. Blood, fecal sampling, and weight data were collected and monitored every 5 days for analysis. No statistical differences were found in the FREE and SEMI groups, but significance was observed in the BARN group. The body weight of the goats gradually reduced from 13.0 ± 2.44 kg to 10.18 ± 2.61 kg (mean ± standard deviation). In contrast, the significantly increasing red blood cells, packed-cell volume, white blood cells, neutrophil-to-lymphocyte (N/L) ratio, cortisol hormone, and antioxidation profiles were observed to be higher in BARN than in FREE and SEMI groups. The intensity of the parasite eggs was remarkably significant. It was observed in the BARN group between the beginning and end of the experiment (supported by a high level of eosinophils). These data can be applied for the realistic evaluation and improvement of management practices, especially fully restrained management (BARN) for monitoring the health status of goats. Effectiveness of raw bacteriocin produced from lactic acid bacteria on biofilm of methicillin-resistant Staphylococcus aureus

Preview Abstract
Probiotics are proven beneficial to health since they enhance immunity against dangerous pathogens and increase resistance to illness. Bacteriocin produced by lactic acid bacteria (LAB), demonstrates a broad inhibitory spectrum and therapeutic potential. This study aimed to isolate LAB-producing bacteriocin and investigate the effect of crude bacteriocin on biofilm from methicillin-resistant Staphylococcus aureus (MRSA). This study used randomly collected 80 white soft local cheeses (40 each from cows and sheep) from different supermarkets in Basrah Province. The obtained samples were cultured and the bacterial suspension of S. aureus was prepared at 1.5 × 108 cells/mL. The crude bacteriocin extracted from LAB was obtained, and the tube was dried and inverted to detect the biofilm loss at the bottom. There were 67 (83.75%) LAB isolates. Among 40 milk samples collected directly and indirectly, there were 36 (83.33%). Staphylococcus aureus isolates based on conventional bacteriological analysis and biochemical tests. Molecular testing was conducted to identify LAB and MRSA. Depending on genotypic results, the effect of white soft local cheese (cows and sheep) and the amplification results of the 16S rRNA gene were detected in 46 LAB isolates from white soft local cheese from cows and sheep. Based on the molecular identification of the mecA, results on Staphylococcus determined that only 2 of 36 isolates of S. aureus carried the mecA. Moreover, there were 26 (86.66%) isolates (MRSA) from samples of raw milk from local markets and subclinical mastitis in cows. The ability of LAB isolates was tested. The effects of bacteriocin production on preventing biofilm growth and formation were investigated. Results demonstrated that bacteriocin has high activity. Microtiter plates applied to investigate the ability of S. aureus to produce biofilms revealed that all isolates were either weak or moderate biofilm producers, with neither non-biofilm nor strong biofilm producers found among the tested isolates. Lactic acid bacteria demonstrate a high ability to produce bacteriocin. Crude bacteriocin from LAB has a restrictive effect on biofilms produced by MRSA; thus, it can be used to reduce the pathogenicity of this bacterium. Transplantation of Aceh cattle ovary into the uterus of pseudopregnant local rabbits: Effect of post-transplant stress on uterine histopathology and ovarian follicle dynamics

Preview Abstract
The increase in the levels of the cortisol hormone caused by the stress conditions generated by an ovary transplantation procedure can damage the uterus of the transplant recipient as well as the transplanted ovaries. This study aimed to analyze the histopathological changes that occur in the uterine horn of pseudopregnant local rabbits (recipients), as well as the ovarian follicular integrity of the donor Aceh cattle after transplantation. After 30 days of adaptation, all rabbits were divided into three treatment groups: R1 (the group of rabbits that underwent ovarian transplantation for 3 days, n = 5), R2 (the group of rabbits that underwent ovarian transplantation for 5 days, n = 5), and R3 (the group of rabbits that underwent ovarian transplantation for 7 days, n = 5). Pseudopregnancy induction was performed using the pregnant mare's serum gonadotropin (PMSG) and human chorionic gonadotropin (hCG) methods. The rabbits were injected with 100 IU of PMSG intramuscularly, followed by an injection of 75 IU of hCG intravenously 3 days later. Ovarian transplantation was performed on day 8 (day 0 was the day of hCG injection). The concentration of cortisol hormone metabolites was measured from fecal samples using an enzyme-linked immunosorbent assay technique. The uterus and ovaries were collected for histopathological and follicular dynamics examination after the transplantation process was completed. The mean cortisol levels (ng/g) recorded before versus after the transplant in the R1, R2, and R3 groups were 146.23 ± 17.60 versus 338.84 ± 302.79, 128.97 ± 81.56 versus 174.79 ± 101.70, and 124.88 ± 43.61 versus 321.91 ± 221.63 (p < 0.05), respectively. The examination of the histopathological appearance of the uterus revealed edema in the uterine lumen, hyperemia and hemorrhage in the endometrium, necrosis of the epithelium, and infiltration of inflammatory cells. Hemorrhage and hyperemia were severe and filled the endometrium in the R1 compared with the R2 and R3 animals. Ovarian follicle development occurred in all treatment groups, although some histopathological features were observed. The number of tertiary follicles in R1, R2, and R3 animals was 24.67 ± 7.37, 20.67 ± 7.57, and 9.67 ± 3.79 (p < 0.05), respectively. Based on the results of this study, it can be concluded that the transplantation of ovaries from Aceh cattle into pseudopregnant local rabbits triggered an increase in the levels of the cortisol hormone and uterine histological changes; however, follicles were still detected at various stages of development in the transplanted Aceh cattle ovaries. The results of this study are valuable for clinicians and researchers because they provide information regarding an alternative in vivo ovarian preservation technique using pseudopregnant rabbits. Distribution analysis of tetracycline resistance genes in Escherichia coli isolated from floor surface and effluent of pig slaughterhouses in Banten Province, Indonesia

Preview Abstract
Slaughterhouses and their effluents could serve as a "hotspot" for the occurrence and distribution of antibiotic-resistant bacteria in the environment. This study aimed to understand the distribution of tetracycline resistance genes in Escherichia coli isolated from the floor surface and effluent samples of pig slaughterhouses in Banten Province, Indonesia. Ten samples, each from floor surface swabs and effluents, were collected from 10 pig slaughterhouses in Banten Province. Escherichia coli strains were isolated and identified by referring to the protocol of the Global Tricycle Surveillance extended-spectrum beta-lactamase E. coli from the WHO (2021). Quantitative real-time polymerase chain reaction (qPCR) was used to detect the tet genes. The tetA, tetB, tetC, tetM, tetO, and tetX genes were distributed in the isolates from the floor surface samples, and the tetA, tetC, tetE, tetM, tetO, and tetX genes were distributed in the isolates from the effluent samples. The tetO gene (60%) was the most dominant gene in the isolates from floor surface samples, while the tetA gene was the dominant one in the isolates from the effluent samples (50%). The tetA + tetO gene combination was the dominant pattern (15%) in the E. coli isolates. The high prevalence and diversity of the tet genes in floor surface and effluent samples from pig slaughterhouses in Banten Province indicated that the transmission of the tet genes had occurred from pigs to the environment; thus, this situation should be considered a serious threat to public health. Growth-stimulating and antioxidant effects of vanillic acid on healthy broiler chickens

Preview Abstract
Phytobiotics are a safe alternative to feed antibiotics in industrial poultry farming that increases productivity by stimulating various digestive enzymes to reduce the number of pathogenic microorganisms in the intestines and improve antioxidant status and immune response. This study aimed to evaluate the effect of vanillic acid in its pure form and in combination with gamma lactone on weight growth and the physiological parameters of broiler chickens. The studies were performed on 120 Arbor Acres broiler chickens (7 days old) that were randomly divided into four groups with five replicates (cages) and six birds per replicate. The control group was fed the basal diet (BD) only. Group I was fed the BD + gamma lactone (average dose 0.07 mL/animal/day). Group II was fed the BD + vanillic acid (average dose 0.07 mL/animal/day). Group III was fed the BD + gamma lactone + vanillic acid in a 1:1 ratio (average dose 0.07 mL/body/day). Live weight of broiler chickens for all group was assessed at the end of each period (age of 7, 14, 21, 28, 35, 42 days). At the end of the experiment (on day 42), 10 broilers with an average live weight were selected for blood sampling from the axillary vein. The results showed a growth-promoting effect of vanillic acid when fed with a diet free of synthetic antioxidants. Groups I and II had increased live weights throughout the entire experiment and a significant increase at the end of the experiment (8.2%–8.5%; p ≤ 0.05) compared to the control group. Toxic effects were not found in the biochemical blood serum analyses of Groups II and III. The metabolic processes in the experimental groups were intensified, especially in the enzyme associated with amino acid metabolism (gamma-glutamyl transferase) in Groups I and III (p ≤ 0.05). Vanillic acid, whether fed alone or in combination with gamma lactone, exhibited high antioxidant activities, protected cells from oxidative damage by inducing total antioxidant, catalase, and superoxide dismutase activities (p ≤ 0.05), and reduced the level of malondialdehyde (p ≤ 0.05) measured. No significant changes in the morphological blood parameters were found. The use of vanillic acid alone and in combination with gamma lactone increases the digestive enzyme activities in blood plasma, increases body weight, and has a positive effect on lipid metabolism and the antioxidant status of broiler chickens. These findings indicate the significant potential use of vanillic acid and gamma lactone in poultry due to their antioxidant properties. Preview Abstract
Gilt selection has the propensity to improve reproductive performance and promote longevity. However, the impact of this process on oxidative stress biomarker levels remains to be unraveled under tropical conditions. This study aimed to determine the effect of management processes during gilt selection on serum malondialdehyde (MDA), catalase (CAT), superoxide dismutase (SOD) levels, and the performance of replacement gilts under tropical environmental conditions. Two groups of 90 crossbred gilts (mean age: 9.72 ± 0.097 weeks) were selected 2 weeks apart, allotted to six pens of 30, and raised in an open barn with shaded roofs. Following their respective entry weeks, gilts in groups one and two were subjected separately to three subsequent selection processes (involving movements, handling, and examination of structural and reproductive traits) at weeks 12, 17, and 24 in the replacement barn. Grower, finisher, and gestation diets were supplied ad libitum for 25 weeks. Environmental temperature (°C), humidity (%), and light (lux) were recorded. Malondialdehyde, CAT, and SOD levels were assayed using blood samples collected on day 1 of experiment (PRE), and at week 24 in replacement barn (POST). Feed intake, weight gain, and percent selected at week 24 were 1.89 versus 1.90 kg/day, 0.81 versus 0.76 kg/day, and 75.23% versus 64.45% for groups one and two, respectively. Sickness, death, slow growth, leg, and reproductive problems caused 24.76% versus 35.55% of removals in groups one and two, respectively. Serum biomarkers were insignificant at PRE but were elevated at POST, with mean values of 14.25 versus 13.84 uM, 5.10 versus 3.26 nmol/min/mL, and p < 0.05, and 1.13 versus 1.68 U/mL and p < 0.05 for MDA, CAT, and SOD in groups one and two, respectively. The impact of the selection process was meager compared to the adverse effect of high environmental temperatures. The management and selection of replacement gilts in an uncontrolled environmental temperature increase the risk of oxidative stress, especially in tropical regions. Staphylococcus aureus and Staphylococcus pseudintermedius are widespread skin and mucous membrane colonizers and may cause opportunistic infections in humans and animals. This study aimed to identify and characterize methicillin-resistant S. aureus (MRSA) and methicillin-resistant S. pseudintermedius (MRSP) isolates from domestic and stray dogs and cats and pet owners in Malaysia using molecular epidemiology and antimicrobial profiling. Three hundred and fifty oral and nasal swabs were taken from pet and stray dogs and cats and pet owners; all samples were subjected to culture and biochemical tests and polymerase chain reaction; the selected isolates were put through disk diffusion test and multilocus sequence typing. One S. aureus isolate and three S. pseudintermedius isolates were identified as MRSA and MRSP, respectively, of which the MRSA isolate and one of the MRSP isolates showed multidrug resistance and the remaining two MRSP isolates were resistant to one or two antimicrobials. Multilocus sequence typing showed that the MRSA isolate belongs to clonal complex (CC) 789, while for the MRSP isolates, two were in CC45 and one was a singleton. This study is the first study in Malaysia to perform molecular characterization of MRSP isolated from pet dogs and cats and pet owners. The outcomes of this study reveal that even healthy pet dogs and cats and their owners can be carriers of drug-resistant staphylococci, highlighting the role of pets and pet owners as carriers of MRSA and MRSP in Malaysia. Preview Abstract
Extensive cattle rearing is a major source of economy for the inhabitants of the Amazon region of Peru. Milk and meat production is generally affected by the prevalence of various parasites, including hepatic and gastrointestinal parasites, as these products provide ideal conditions for parasitic growth. This poses a serious public health threat. This study aimed to estimate the prevalence, coinfection, and risk factors associated with the liver fluke (Fasciola hepatica) and other gastrointestinal parasites in cattle from the Amazon region of Peru. Fecal samples obtained from 1450 bovine specimens were analyzed using flotation and sedimentation methods to identify parasites, including Eimeria spp., strongyle-type eggs (STEs), and F. hepatica. We collected information about the specimens, including age, sex, origin, breed, category, frequency of deworming, farm size, herd size, water sources, and rearing system by conducting simple inspections and interviewing owners. The data obtained were statistically evaluated using the Chi-square test (p < 0.05) to determine the association between the qualitative variables. We also calculated the odds ratio at a 95% confidence interval to identify the risk factors. We observed that F. hepatica, Eimeria spp., and STEs were 45.6%, 39.8%, and 35.3% prevalent, respectively. We found risk factors related to distomatosis in the animals from Huambo, where the drinking water sources are mainly streams, ditches, and rivers, while the specimens from Valle Chico were predisposed to coccidiosis. Further, the risk factors related to the presence of STEs in feces were age (61–90 months), origin (Valle Chico), herd size (<50 animals), and type of extensive rearing. Furthermore, significant coinfection was observed between Eimeria spp. and STEs. The high percentages of parasites in cattle observed were related to epidemiological factors, such as the origin of the sample, water sources, age, herd size, and extensive breeding. Similarly, the presence of STEs was a risk factor for contracting coccidiosis. Our future goals include investigating these parasites using a larger sample size and identifying more risk factors using more sensitive and specific diagnostic tests. The improper handling of poultry litter and waste poses risks to humans and environment by introducing certain compounds, elements, and pathogenic microorganisms into the surrounding environment and food chain. However, understanding the farmers' knowledge, attitude, and practices (KAP) could provide insights into the constraints that hinder the appropriate adoption of waste management. Therefore, this study aimed to assess poultry farmers' KAP regarding waste management issues. A cross-sectional KAP study was conducted with native poultry keepers and small-scale commercial poultry farmers in seven districts of Bangladesh. In the survey, 385 poultry producers were interviewed using validated structured questionnaires through face-to-face interviews to collect the quantitative data in their domiciles. The overall KAP of farmers regarding poultry waste management issues demonstrated a low level of KAP (p = 0.001). The analysis shows that roughly 5% of farmers have a high level of knowledge of poultry waste management issues, followed by around one-third of respondents having a moderate level of knowledge. Considering the attitude domain, more than one-fifth of native poultry keepers and nearly two-thirds of commercial producers demonstrated a low level of attitude toward poultry waste management. Considering the overall analysis, roughly half of the respondents found a high level of attitude, and over half of the farmers showed a moderate level of attitude toward poultry waste management issues. The analysis showed that the level of good practices for native and commercial poultry production systems is estimated at 77.3% versus 45.9%, respectively, despite the farmers' lesser knowledge and attitudes toward poultry waste management systems. Overall, analysis showed that nearly 60% and 40% of poultry producers had high and moderate levels, respectively, of good practices in poultry waste management issues. Analysis of the KAP data shows that farmers had a low level of KAP toward poultry waste management. The result of this study will assist in formulating appropriate strategies and to adopt poultry waste management solutions by poultry farmers to reduce environmental degradation. Intestinal infections are associated with Gram-negative bacteria like Escherichia coli. When eliminated by treatments during replication, E. coli release lipopolysaccharides (LPS) that can activate the intestinal immune system and increase the expression of cytokines, such as interleukin (IL)-8, IL-18, tumor necrosis factor-alpha (TNF-α), and interferon-gamma (IFN-γ), by the intestinal epithelium under pathological conditions. This study aimed to evaluate the addition of Bacillus subtilis to the duodenal gene expression of pro-inflammatory and anti-inflammatory cytokines in broilers exposed to LPS from E. coli. RNA was extracted using the Zymo Research total RNA commercial kit, according to the manufacturer's specifications, from the intestinal tissue of the duodenum previously resuspended in the lysis buffer of the kit. The expression of the cytokines of interest was measured using the QuantiNova SYBR green real-time polymerase chain reaction kit (Qiagen). Transcript quantification was performed by the ΔΔC(t) method using glyceraldehyde 3-phosphate dehydrogenase as a normalizing constitutive gene. For the measurement of pro-inflammatory (IL-8, IL-18, TNF-α, and IFN-γ) and anti-inflammatory (IL-10) cytokines, there was no statistically significant difference (p > 0.05) between the basal diet and the diet with antibiotic (avilamycin). There was a statistical difference (p < 0.05) between diets with LPS. The diet with B. subtilis presented the lowest expression; the results differed on each sampling day (days 14, 28, and 42). A decrease in the expression of pro-inflammatory cytokines (IL-8, IL-18, TNF-α, and IFN-γ) and an increase in IL-10 (anti-inflammatory) was observed; in this way, a balance of the inflammatory response to bacterial infection is achieved, suggesting that the use of B. subtilis as an additive in a broiler diet has a similar effect to that produced with antibiotic growth promoter. Mycological evaluation of frozen meat with special reference to yeasts

Fungi can play beneficial and detrimental roles in meat products; however, the diversity and significance of fungi in meat products are poorly understood. This study aimed to isolate and characterize fungal species from frozen beef samples collected from retail stores in the Qena Governorate, Egypt. A total of 70 frozen beef samples were collected from retail stores in Qena, Egypt. All samples were subjected to mycological examination. Fungal colonies were identified using conventional approaches, as well as the VITEK 2 system and DNA sequencing of the internal transcribed spacer region. Analyses of enzymatic activity, biofilm formation ability, and the antimicrobial resistance profiles of the isolated yeasts were also conducted. Molds and yeasts were isolated from 40% and 60% of meat samples, respectively. Mold isolates were dominated by Aspergillus, Penicillium, and Cladosporium spp., whereas yeast isolates were identified as Candida albicans, Candida parapsilosis, Yarrowia lipolytica, Saccharomyces cerevisiae, and Rhodotorula mucilaginosa. Compared to other yeast species, the highest production of lipase and protease was observed in Candida species. The strongest ability to form biofilms was observed in Candida spp., followed by S. cerevisiae, Y. lipolytica, and R. mucilaginosa. The results of antimicrobial susceptibility testing revealed that all yeast isolates showed notable resistance to fluconazole and itraconazole. A significant correlation between antimicrobial resistance and biofilm formation was observed in several species. This study highlights the importance of the dangers of yeasts in food products and the extent of their impact on public health. Bovine trichomoniasis, caused by Tritrichomonas foetus, is a venereal disease that is distributed in many countries, including Iraq. Compared with other abortive infectious diseases, prevalence of T. foetus is expected to be relatively low in the field by veterinarians. This study aimed to estimate the prevalence of T. foetus in aborted cows by conventional polymerase chain reaction (PCR) and phylogenetic analysis of local T. foetus isolates was documented in the National Center for Biotechnology Information as the first sequenced isolates from Iraq. Vaginal fluids were collected from 62 aborted cows and examined by PCR. Data were reported for the following parameters: Vital signs (body temperature and respiratory and pulse rates), age (<4, 4–8, and >8 years), reproductive health status (premature calving, embryonic death, pyometra, and healthy newborn), breed (pure or crossbred), type of breeding (natural or artificial), bull-to-cow ratio (1:<10, 1:10–20, and 1:>20), contact of cow with bull(s) from other farmers (yes or no), and contact with stray animals (dogs and cats). A total of 20.97% of aborted cows were positive for T. foetus. Phylogenetic analysis for 10 positive local T. foetus isolates demonstrated high identity with the Thai (MN560972.2) and Chinese (MH115435.1) isolates, with an identity range of 98.8%–99.5% and 98.6%–99.3%, respectively. Clinical data showed that the vital signs differed insignificantly between cows positive and negative for T. foetus. Prevalence and risk of infection increased significantly in <4-year-old, early calving, embryonic death, crossbred, and naturally inseminated cows that had direct contact with bulls from other farmers, and contact with stray animals. Fetal pneumonia and death of premature calves were significant among positive aborted fetuses. Tritrichomonas foetus is highly prevalent in aborted cows in Iraq and phylogenetic analysis demonstrated an identity between the local and global isolates, that is, Thai and Chinese, of cats. The use of alternative feeds in feeding livestock as an alternative to traditional feeds has been used for many years, on the one hand, to lower the price of feed and, on the other hand, to raise the profitability of raising livestock. The study aimed to investigate the effect of feeding pomegranate seed pulp (PSP) on the growth performance and carcass characteristics and the health of Awassi lambs. Twenty-four male lambs (16.9 ± 0.42 kg) were assigned randomly to one of two isonitrogenous (160 g/kg crude protein of dietary dry matter [DM]) treatment diets. The diets were the control (CON) and PSP-containing diet (100 g/kg of dietary DM; PSP100). The experimental period was 70 days preceded by 7 days of adaptation to diets. Feed intake was measured and lambs were weighed on day one and then biweekly. On day 49, eight lambs (four lambs per treatment) were chosen randomly and placed in metabolic cages for a digestibility trial. At the end of the trial, lambs were slaughtered to evaluate carcass characteristics and meat quality. The data were analyzed using Proc Mixed procedures of SAS. The results revealed that nutrient intake was greater (p < 0.05) in lambs fed PSP100 than those fed the CON diet. Nitrogen intake and nitrogen retention were greater (p < 0.05) for lambs who consumed the PSP100 diet compared to CON. Final weight, total gain, and average daily gain were greater (p < 0.05) with lambs fed PSP100. Hot and cold carcass weights were higher (p < 0.05) by the PSP100 group than by the CON group. Carcass cut weight increased (p < 0.05) with feeding PSP100 diet. No differences were detected in blood parameters except high-density lipoprotein content, which was greater (p < 0.05) in the PSP100 group compared with the CON group. It could be concluded that adding PSP to lambs' diets improved growth and carcass measurements positively and did not negatively affect lambs' health; therefore, it is recommended to use PSP as an alternative to traditional feeds in lambs formulated rations. Chronic unpredictable mild stress (CUMS) is a protocol widely used to create an animal model of depression with food deprivation, water deprivation, and physical-dominant stressors as routine procedures. However, human depression mainly involves psychological stressors and does not always involve a lack of food and water; thus, CUMS procedures should be modified accordingly. Therefore, this study aimed to create an animal model of depression, mainly focusing on a psychologically dominant stressor without food and water deprivation. The CUMS and control groups, respectively, received CUMS modification (psychologically dominant stressors without food and water deprivation) for 21 days. A 24-h sucrose preference test (SPT) was used to assess the successful creation of an animal model of depression. Daily food intake measurements, weekly weight monitoring, and weight gain calculations were performed. Either an independent sample t-test or the Mann–Whitney test was used. Of the 42 rats included, 39 completed the study. Chronic unpredictable mild stress procedures for 21 days significantly reduced the SPT (p < 0.05), mean body weight (p < 0.05), and weekly weight gain (p < 0.05) in the CUMS group compared to the control group. However, the weekly average food intake did not statistically differ between the two groups. Psychological dominant CUMS modification to an animal model of depression resulted in lower SPT, body weight, and weekly weight gain in the CUMS group than in the control group. Heat shock proteins are highly conserved proteins that work as molecular chaperones expressed in response to thermal stress. This study aimed to determine the expression profile of genes related to the heat stress response in whole blood obtained from the Romosinuano creole breed. Real-time polymerase chain reaction was performed to analyze the transcript of hsp90, hsp70, hsp60, and hsf1 in the whole blood of Romosinuano under different temperature-humidity indices (THIs). The expression levels of the hsp70 and hsf1 genes at the high-THI level were higher (p = 0.0011 and p = 0.0003, respectively) than those at the low-THI level. In addition, no differences in the expression levels of the hsp60 and hsP90 genes were detected between the two THIs. The overexpression of hsf1 and hsp70 genes play an important role in protecting cells from damage induced by heat stress. Biosecurity implementation is fundamental to combating diseases and antibiotic resistance. Therefore, this study aimed to examine the correlation between the implementation of biosecurity measures in small-scale duck farms and the incidence of infectious diseases that threaten the duck industry. Twenty small-scale duck farms of different breeds and production stages were collected as representative samples, focused on two districts in the Qalyoubia governorate, which possesses high-density small-scale farms. A 30-point structured questionnaire was designed to assess the level of biosecurity measures implemented in the sampled farms. These farms were examined for bacterial infection by cultivation, typing, and antibiotic sensitivity tests, in addition to molecular techniques for detecting suspected viral diseases. The results showed that the farms had high or low levels of biosecurity; only 25% possessed high-level biosecurity. Bacteria, including Salmonella, Escherichia coli, Staphylococcus, and Pasteurella, were isolated from all sampled farms. High rates of antimicrobial resistance-reaching up to 100% were observed against some drugs. However, viral causative agents, including HPAI-H5N8, duck viral hepatitis, and goose parvovirus, were isolated from only five farms. The lack of commitment to biosecurity implementation, particularly personal hygiene, was observed in most sampled farms. Increasing the level of biosecurity reduced the incidence of mixed infections. Feline infectious peritonitis (FIP), one of the most important infectious diseases in cats is caused by FIP virus (FIPV), a mutated variant of feline coronavirus. Feline infectious peritonitis has a negative impact on feline health, with extremely high mortality in clinical FIP-infected cats, particularly young cats. There are no approved drugs for FIP treatment, and therapeutic possibilities for FIP treatment are limited. This study aimed to utilize nature-derived bioactive flavonoids with antiviral properties to inhibit FIPV infection in Crandell–Rees feline kidney (CRFK) cells. The cytotoxicity of 16 flavonoids was evaluated on CRFK cells using a colorimetric method (MTS) assay. Viral kinetics of FIPV at 50 tissue culture infectious dose (TCID50)/well was determined during the first 24-h post-infection (HPI). Antiviral activity was evaluated based on the replication steps of the virus life cycle, including pre-compound, attachment, penetration, post-viral entry, and virucidal assays. The antiviral efficacy of flavonoids against FIPV was determined based on positive FIPV-infected cells with the immunoperoxidase monolayer assay and viral load quantification using reverse transcription-quantitative polymerase chain reaction. Two flavonoids, namely, isoginkgetin and luteolin, inhibited FIPV replication during post-viral entry in a dose-dependent manner, with 50% maximal effective concentrations = 4.77 ± 0.09 and 36.28 ± 0.03 μM, respectively. Based on viral kinetics, both flavonoids could inhibit FIPV replication at the early stage of infection at 0–6-HPI for isoginkgetin and 2–6-HPI for luteolin using a time-of-addition assay. Isoginkgetin exerted a direct virucidal effect that reduced the viral titers by 2 and 1.89 log10 TCID50/mL at 60 and 120 min, respectively. Isoginkgetin interfered with FIPV replication during both post-viral infection and virucidal experiments on CRFK cells, whereas luteolin inhibited the virus after infection. The reproductive success of Simmental bovine after sex-sorting under various incubation and centrifugation protocols

Preview Abstract
To enhance the reproductive potential and increase productivity and population of cows, spermatozoa sex-sorting technology is required. This study aimed to examine the effect of sexing sperm, separated using a bovine serum albumin (BSA) column with varying incubation durations and centrifugation methods, for successful artificial insemination. Six Simmental bulls and 30 cows (n = 30) as the recipients were selected for this study at Balai Pembibitaan Hijauan Pakan Ternak Sembawa Indonesia. The study parameters included sperm motility, viability, plasma membrane integrity, and conception rate (CR). The experiment was divided into three protocols to find out differences in some parameters: (1) BSA incubation time effect (P) with P1 (40 min), P2 (50 min), and P3 (60 min); (2) freezing time effect with before freezing and after-thawing treatments; and (3) CR determined by measuring the proportion of pregnant cows following insemination with non-sexed, X-bearing, and Y-bearing sperms without centrifugation (n = 15) (A0, A1, and A2) and with centrifugation (n = 15) (B0, B1, and B2) in the acquired data, which were counted using the Statistical Package for the Social Sciences version 21 program. Analysis of variance was utilized to evaluate all treatments at various levels. The results demonstrated that centrifugation time influenced all sperm quality metrics for sperm containing X and Y (p < 0.05). The non-return rate (NRR) of non-sexed frozen semen, both centrifuged (A0) and not centrifuged (B0), was more significant than frozen semen produced by sexing X and Y spermatozoa. The NRR indicated a value of 80% based on the number of lactating cows. Bovine serum albumin incubation and centrifugation protocols influenced and decreased all sperm quality indicators throughout the sexing procedure and could still be used as a sexing protocol. Vannamei shrimp (Litopenaeus vannamei) is an important food commodity of economic benefit due to its high price, low susceptibility to disease, and popularity for consumption. These advantages have led many farmers to cultivate vannamei shrimp. Efforts are underway to improve the aquaculture performance of this species, including the use of probiotics, which are non-pathogenic bacteria that aid in digestion and help fight disease. Probiotics are usually obtained from the intestines of vannamei shrimp or the culture environment. They are low-cost, non-pathogenic, and largely non-toxic source of antibiotics and are able to synthesize various metabolites that have antibacterial functions and applications. Research on probiotic use has primarily been focused on increasing vannamei shrimp aquaculture production. Bacterial species, such as Lactobacillus or Nitrobacter, can be administered orally, by injection, or as a supplement in aquaculture water. Probiotics help to improve survival rate, water quality, immunity, and disease resistance through space competition with disease-causing bacteria, such as Vibrio spp. An increased number of probiotic bacteria suppresses the growth and presence of pathogenic bacteria, which lowers disease susceptibility. In addition, probiotic bacteria also aid digestion by breaking down complex compounds into simpler substances that the body can absorb more easily. This mechanism improves growth performance in terms of weight, length, and feed conversion ratio. This review aimed to provide information regarding contribution of probiotic to improve vannamei shrimp production in aquaculture.

Preview Abstract
In several countries, two Entamoeba porcine species, Entamoeba suis and Entamoeba polecki (subtype 1 and 3), have been detected using molecular methods and identified pathogenicity associated with enteritis. However, globally, Entamoeba infection prevalence in pigs is extremely limited. This study aimed to coprologically and genetically examine pig parasites to estimate prevalence of Entamoeba in three pig farms in East Java, Indonesia. Hundred porcine fecal samples (Landrace) were collected from three East Javan farms in well-known swine industry regions. Fecal samples were examined under microscope after sugar-flotation centrifugation, and molecular species and subtype identification were performed using polymerase chain reaction (PCR) and primer pairs targeting small-subunit ribosomal RNA. Microscopy examinations identified parasites in 89/100 fecal samples; Entamoeba spp. cysts were the most frequent in these samples. Polymerase chain reaction showed that 58 samples were comprised of mixed Entamoeba suis and Entamoeba polecki, 22 E. suis alone, and nine E. polecki alone infections. Epolec F6–Epolec R6 primers successfully amplified E. polecki ST1–4 subtypes, while Epolecki 1–Epolecki 2 amplified only the E. polecki ST1 subtype. Entamoeba polecki ST1-specific primers successfully detected the ST1 subtype in 19/67 E. polecki positive samples. Entamoeba spp. prevalence in Indonesian pigs was previously shown to be high. On coprological examination of East Javan pigs, we detected high Entamoeba spp. levels, in which we genetically identified as E. suis (80.0%), E. polecki (67.0%), and E. polecki ST1 (19%). Prevalence and resistance to antibacterial agents in Salmonella enterica strains isolated from poultry products in Northern Kazakhstan

Salmonella is one of the main causative agents of foodborne infections. The source of the pathogen, in most cases, is poultry products. The intensification of poultry farming and the constant and uncontrolled use of antimicrobials has led to an increase in the level of antibiotic resistance, especially in developing countries. This study aimed to determine the level of sensitivity to antimicrobial agents in Salmonella enterica strains isolated from poultry products in Northern Kazakhstan, as well as to determine the genetic mechanisms of resistance and the presence of integrons. In total, 398 samples of poultry products sold in Northern Kazakhstan were selected. Salmonella strains were isolated from product samples using microbiological methods. Salmonella was identified based on morphological, biochemical, and serological methods, as well as polymerase chain reaction (PCR). Sensitivity testing for antimicrobial agents was performed using the disk diffusion method. The detection of resistance genes was performed using PCR and gel electrophoresis. Out of 398 samples of poultry products, a total of 46 Salmonella isolates were obtained. Most of the isolates belong to the serovar Salmonella Enteritidis (80.4%). The assessment of sensitivity to antibacterial agents showed that Salmonella was mainly resistant to nalidixic acid (63%), furadonin (60.9%), ofloxacin (45.6%), and tetracycline (39.1%). In 64.3% of cases, Salmonella was resistant to three or more groups of antibacterial agents. Resistance genes such as tetA, tetB, blaTEM, aadA, sul3, and catII, as well as integrons of two classes (teg1 and teg2), were identified. Poultry products contain antimicrobial-resistant strains of Salmonella, as well as genes encoding resistance mechanisms. The results emphasize the need for constant monitoring of not only pathogenic microorganisms but also their sensitivity to antimicrobial agents. The potential threat to human health requires a unified approach to the problem of antibiotic resistance from representatives of both public health and the agroindustrial complex.
